Circulating Tumor Cell

The SKCC Circulating Tumor Cell (CTC) Shared Resource Core Facility provides services on CTC enrichment and enumeration based on the Food and Drug Administration (FDA)-approved CellSearch™ platform, and single-CTC isolation based on the DEPArray™ platform. The CellSearch™ platform is able to enrich and enumerate CTCs from 7.5 ml whole blood of patients of most epithelial cell-derived cancers (e.g., breast, prostate, colorectal, lung, head and neck, skin, and other cancers). CTCs are cancer cells that detach from solid tumors and enter into blood stream. CTC enumeration and single-cell isolation may be performed prior to the start of therapy and during the course of treatment. The isolated single CTCs can be subjected to various molecular and genomic characterizations such as next-generation sequencing in order to identify CTC-based biomarkers that inform the precision treatment of cancer patients. Serial analyses of CTC enumeration and single-cell analysis could be used in conjunction with other clinical methods for longitudinal and non-invasive monitoring of the treatment response and disease progression of cancer patients.

Services

CELLSEARCH®-based CTC enumeration

CELLSEARCH® CTC Test is the first and only clinically validated, FDA-cleared blood test for enumerating CTCs. This “Liquid Biopsy” test can be used at any time during a patient's course of disease. Using a simple blood draw, the CELLSEARCH® Circulating Tumor Cell assay is intended for the identification of CTCs of epithelial origin (EpCAM+, Cytokeratins 8, 18+, or 19+, and CD45-) in 7.5 mL of whole blood sample prepared by CellSave Preservative Tubes within 4 days. CELLSEARCH® Circulating Tumor Cell Kit was designed for use with the CELLTRACKS® AUTOPREP® System and the CELLTRACKS ANALYZER II® System.

DEPArray™ automated platform for single-cell isolation with high fidelity recovery

Dielectrophoresis (DEP) cages of the platform allow the trapping, manipulation, and recovery of individual cells on the DEPArray™ System for multiple cells and cell types in a heterogeneous sample for further downstream molecular applications, including cell culture, single-cell whole genome amplification and next-generation sequencing.

The CTC core facility has established a complete pipeline on blood processing, CTC enrichment and enumeration, single-CTC isolation, whole genome amplification, whole-exome and targeted next generation sequencing, and data analysis. The core will work with researchers and clinicians to provide assistance to incorporate CTC-related components into their research applications and clinical protocols.

Whole blood sample preparation and submission:

The hardcopy of the request form must have a valid charge code and be approved by the Prinicipal Investigator for billing.

All Samples must be whole blood, at least 8 mL and be stored in CellSave test tubes.

Please provide a label (required first), type of cancer, stage of cancer, any treatments, collection date and time for each sample.

CELLSEARCH® and DEPArray™ CTC identification request forms with relevant materials should be delivered to Room 727 in College Building.
